Why is the poison mushroom a big deal, exactly? It's pretty easy to identify, what with the purple smog around it.

Possibly because it was unexpected? In any case, that it appears in what seems to be the first Special World level shows that the game is taking of the kid gloves there.

Hm, and people say Sonic is a chatterbox...
Being a voice actor for Mario normally isn't very labor-intensive, normally the longest sentence he says is "thank you so much for playing my game!" after the end credits.
Doing promotional videos (the one for Mario Sports Mix where Mario was a coach, for example) is a good way to get more out of the voice, Charles Martinet does a really good job with it.
Yeah, there was another vid that had him doing a mock interview with a Wii host (or was it a puppet), that just like this one made me surprised at Martinet having Mario speak full sentences that weren't in catchphrase-ese.
Not that I'd have him voice the character if I were to make an animated show or something.

well Mario's voice is more of a cartoon character like Mickey mouse than say solid snake.
If I recall, Mario had some extensive voice work for the first Mario Vs Donkey Kong.
