- Defictionalization: Since the film's release, A marijuana strain named Pineapple Express was bred from crossing Trainwreck and Hawaiian
- Hilarious in Hindsight:
- Amber Heard being shocked that someone would call her a lesbian. It was probably more an Actor Allusion because even though she wasn't out, the cast (including writer Seth Rogen) likely knew.
- Angie's May–December Romance with Dale is pretty funny considering that Amber Heard was once married to the much, much older Johnny Depp.
- "If marijuana is not legal within the next five years, I have no faith left in humanity—period!" Sure enough, Washington and Colorado became the first US states to legalize recreational marijuana use in 2012, just four years after the film came out which also makes this Heartwarming in Hindsight.
- Ho Yay: "I'm gonna save you, man!"
- One-Scene Wonder. Bill Hader plays a US Army soldier given marijuana to see how it effects him. Hilarity Ensues.
- Retroactive Recognition: Ken Jeong has a small role in the film as a Mook Lieutenant who leads the attack on Ted's farm compound. He gets to say a few lines and plant the bomb that kills Ted.
- Squick: Dale trying to put part of his ear back on after getting a piece of it shot off by a gun.
- This Is Your Premise on Drugs: Hot Fuzz or Die Hard on weed.
