     Pre-Release 
Madame Web will witness another universe of Spider-Man and will meet him in the future.
A version of Madame Web made an appearance in Spider-Man: No Way Home as one of the visitors after cracks happened thanks to Raimiverse Green Goblin broke up Machina de Kadavus box that had the visitors in it.

So it definitely makes sense for her to notice and witness it in The Stinger to happen for her to highly meet the MCU Spider-Man in the future.

Julia, Mattie and Anya are supervillains.
Ezekiel's comment in the trailer about how Madame Web “doesn't know what those girls will become” implies the situation is more complex than it appears while later Cassandra mentions that their futures “were almost so different”, and in the future Ezekiel's seen they become supervillains. Cassandra will decide to protect them anyway and this will lead them to become heroes instead of villains.
  • Jossed, the girls are destined to become superheroes and Ezekiel is a villain they were trying to stop.

The movie will take place in the past and not in the present.
According to what could be seen on the set of Madame Web, it seems that the film will take place in the past, more specifically in the 2000s and not in the present. Now it will depend if new trailers show smartphones or other modern devices that deny this.
  • Confirmed, the film's main story is set in 2003.

At some point during the final climax, Madame Meb will go blind for life.
It is likely that during the final battle against Ezekiel Sims, Cassandra will lose her vision and go blind forever, forcing her to wear dark glasses or a long scarf to cover her eyes. That would lead to Madame Web looking a little more like her original comic book counterpart.
  • Confirmed, in the film's climax Cassie is rendered blind and paralysed, requiring her to use an electric wheelchair and cover her eyes with dark glasses.

The movie is, alongside Spider-Man: Spider-Verse, teasing that Morlun will be a threat in both cinematic universes.

The movie isn't set within the main Sony Spider-Man universe… yet.
But the events of the film will create the divergent timeline that Venom, Morbius, and Kraven exist in. It may even explain why so many figures associated with Spider-Man are present but the actual hero has yet to be seen.

Jonathan Webb is going to appear too.
Perhaps, he will appear as a future Love Interest for Cassandra Webb.

Ezekiel is hunting potential Spiders to elude the Inheritors.
Like the comics, his powers weren't earned and he isn't a real Spider Totem. This lets him fly under the radar, so long as he kills anyone with the potential to become a true Spider Totem. He fears that all four heroes gaining their powers at once will be like sending up a signal flare for the Inheritors.
  • Seemingly Josssd, Ezekiel makes no mention of the Inheritors. His motivation for hunting down the future Spider-heroines is because he has prophetic dreams of them killing him in the future.

     Post-Release 
The movie's version of Peter Parker is the MCU Spider-Man
We never see May who Ben has recently started dating and we never knew what Peter's parents or Uncle Ben (presuming he exists in the MCU) looked like.
  • Not possible. MCU Peter was born in 2001, not 2003.

Cassie was driving the truck that took out Olivia Octavius.
Car Fu seems to be her favorite tactic. (Don't take this too seriously, it's just for fun).
