The Marvel Cinematic Universe will be retconed to be Earth-001
Just like the Arrowverse. All previous live-action non-MCU Marvel movies and TV shows will be part of the MCU's multiverse.

Well, from what we have seen from the silhouettes in No Way Home, they do have various villain members of Spider-Man, since Strange said that The Visitors have Spider-Man’s Allies

The Marvel Cinematic universe version of Exiles, though taking cues from the 2018 version and not solely be using X-Men characters. The group would consist of characters from non-MCU films and shows coming together as the Exiles to do the job of keeping the multiverse safe. Not all are heroes though, as they would need to take a life if needed to do the job. Members could include: Jamie Chung's Blink, Jennifer Garner's Elektra, Colin Farrel's Bullseye, post-No Way Home Doc Ock, Olivia Munn's Psylocke, Logan-verse's X-23, pre-reboot Fantastic Four's Thing, etc.

Avengers: Secret Wars will partially adapt 2015's Secret Wars, and have the Multiverse destroyed and reforged into 1 single universe
This would allow the MCU to receive a "light reboot, allowing past characters (Iron Man, Natasha Romanoff, Steve Rogers' Captain America) to be rebooted in new franchises, but would keep surviving existing characters intact as they are. Mysterio knew/knows something we don't
Depending on if he really did survive Far From Home. Mysterio is a dangerous foe for sure. But he's never (to my knowledge) been a multiversal level threat (unless you count Shattered Dimensions). Now, we know he's full of shit and isn't actually from a different Earth. But there might be some truth to what he was saying. Think about it, you don't just pull multiversal knowledge like that out of your ass. Name dropping a specific Earth? I think he knows more than he was letting on.
