    Pre-release theories 
Dru will not appear in the film because he returned to the mansion he lived in the third film.
  • Or alternatively, he will appear on the film, either working for the villains or already having been arrested by the AVL.
Poppy’s backstory.
Poppy was part of the orphanage Margo, Edith and Agnes were adopted from by Gru in the first movie, who became an aspiring supervillain after never being adopted. Poppy will be revealed to be Maxime and Valentina’s daughter who got sent to the orphanage for unknown reasons.
The Japanese Title
The Japanese title of this movie is "Minion Super Transformation". This could imply the movie will feature the Minions getting their own transformer style mech (since a Minion turning into a giant had already been done before) to fight off the Big Bad, possibly as the movie's climax.
  • Jossed for now. The second trailer reveals the Minions being the first trials of the AVL’s super-serum, becoming the super-powered Mega-Minions.
The Mega-Minions are Advertised Extras.
While these new superhero forms are prominent in the second trailer, their transformations are the only sequence shown in said trailer with no real showing of them actually utilizing their powers for heroic purposes. Not to mention every other scene with the Minions in both trailers and the TV spots have them in normal forms (but then again, only five Minions "volunteered" to try the serum). So either Illumination for some reason wants to keep the Mega-Minions' real scenes hidden before the movie's release, or maybe there's just not that many scenes of them to show that aren't the third act and Illumination might not want to show them (then again, they are infamous for spoiling the final acts of many of their films in the past, but no scene of any of the two trailers for this film seems to logically allude to the climax of the finished film).

So my theory is, that the Mega-Minions actually don't appear in the final film until the third act, when Gru and/or his family are in their Darkest Hour, so the AVL decides to finally put the super-serum to the test with the Minions, and then the Mega-Minions leap in for the film's final battle. I'm just not convinced that the Mega-Minions will actually have that much screentime in the film, even with all of the marketing and toys focusing just on them. We might just be seeing a Transformers: Age of Extinction Dinobots situation in our hands again.
