  • This line from the trailer, which may double a Moment of Awesome:
    Sam: It's not over until I teabag every last one of you alien motherfuckers.
  • The Serious Sam BFE Helpline promotional videos.
    Rich Knuckles: You can stand there all you want, Mr. Elliot, but your health isn't gonna automatically regenerate. Stop being such a f[BEEP]ing p[BEEP]y.
  • Sam's first encounter with a Beheaded Kamikaze, which mirrors a similar occurence in TFE:
    Headless Kamikaze: AAAAAAAAAAAAAAAAAAAAAAAAAAAAAAAHH!!!
    [Sam kills the Headless Kamikaze'']
    Sam: AAAAAAAAAAH— put a sock in it.
    [Hordes of Headless Kamikazes approach]
    Hordes of Headless Kamikaze: AAAAAAAAAAAAAAAAAAAAAAAAAHHHHHHH!!!
    Sam: Uh oh.
  • The cutscene of the second level, "Into the Spider's Nest", where Sam locates Professor Stein's phone next to his severed arm. Quinn had been calling, and when Sam responds, telling her that Stein is dead, she asks if he found his phone. Adding to the funny, even the music stops for a double take.
    Sam: (in a "are you fucking kidding me" tone) You mean the phone you just called??
    Quinn: Uh... right.
  • Sam exploding a bridge in "Under the Iron Cloud", complete with him whistling the Boy Scouts song:
    Sam: Nothing says 'honey I'm home' like a stick of dynamites.
    [Sam detonates the explosives.]
    Quinn: Jesus Christ, Sam! Do you have to blow EVERYTHING up?
    Sam: As a matter of fact, I do.
  • Sam's first encounter with a Witch-Harpy near the end of "Under the Iron Cloud":
    Sam: Was your daddy a pilot? Because you are...
    (Harpy opens her mouth)
    Sam: ...really freakin' ugly!!
  • Everything surrounding Sam's blowing up of The Sphinx in the level "The Silent Riddler", especially his ensuing argument with Quinn.
    • It all starts at the end of the previous level, "Under the Iron Cloud".
    Quinn: The scientists were able to decipher a map. Apparently there's another Sirian chamber beneath the great pyramid. (...) Here's the weird thing. There's no direct way in. The map just shows this isolated bubble. The words on it say 'the sphinx is the key.' Whatever that means.
    Sam: I think it means the Sphinx is the key. Now, if you'll excuse me, I'm off to blow up a big stone kitty.
    Quinn: Sam, the scientists are almost positive we can figure out a solution.
    Sam: Tell 'em I already thought of a one.
    Quinn: Sam, you can't blow it up, it's a priceless artifact.
    Sam: You'll just have to bill me.
    Quinn: Sam, this is ridiculous, you can't...
    (Sam cuts the call)
    • Then, after he reaches the Sphinx, makes it clear to HQ that his patience is running low:
    Sam: Quinn, I've reached the sphinx. Tell me you have this solved.
    Quinn: Not just yet. We think we're close though.
    Sam: Close doesn't do me much good. Sounds like it's time for plan B.
    Quinn: Sam, for God's sake, it's one of Earth's oldest artifacts...
    Sam: OK, so I'll blow it the hell up really careful.
    Quinn: Do you want to undo all the work we've done?
    Sam: You keep looking for answers. I'll keep looking for dynamite.
    • And after he set the last sticks of dynamite around it:
    Sam: Quinn, your eggheads solve that riddle yet? Plan B is good to go.
    Quinn: Just a few more minutes.
    Sam: Take your time.
    (Sam cuts the call)
    Sam: Time for plan B.
    (Dynamite sets off, and off goes the Sphinx.)
  • After activating the second generator and seeing his escape route being destroyed, Sam screams "Jesus Harold Faltermeyer Christ!"
  • The Final Boss fight, with Sam walking in on Ugh-Zan IV throwing down with the Sand Whale that had been chasing him.
    Sam: You guys having an ugly contest? I'll make it easy for you, you both win!
    • And shortly thereafter, in a call with Mental's daughter as Sam threatens to kill Mental:
      Mental's Daughter: Are you serious?
      Sam: Why yes, as a matter of fact I am.
    • And then they manage to turn the destruction of the Earth into a mooning joke.
  • From the Jewel of the Nile DLC we have this gem:
