Since they are going to be on their journey to the multiverse, it is likely they will be recruiting heroes from the DC Multiverse to stop Lex and his Legion of Doom. Harleen is going to turn against the Legion of Doom
Given that she is shown to be with Babs and her friends, it is heavily implied that she will be betraying the Legion of Doom.- Confirmed

Since it was said that Lex Luthor united the villains of the world to form the Legion of Doom, it is implied that the villains from the Multiverse will be joining him as well.- Jossed

If you look closely in the trailer, you will notice, you will see that her hair is silver, and heart of her clothing is blue.- Confirmed
Cythonna and DCSHG Lex will serve as the Big Bad Duumvirate
Since they are going to be teaming up together while serving as leaders of the Legion of Doom, it is implied that they will presumably try to take over the Multiverse. DCSHG Lex will end up with a Karma Houdini Warranty
Well, since he’s one of the only villains not to be punished, besides his stuff being taking away by Catwoman, he might end up being betrayed, and exposed for his crimes.- Confirmed

The SHG will finally realize the Super Villain Girls’ secret identities
Diana and Barbara, we're the only ones that realize Harleen is Harley Quinn, except for the rest of the five. So, it is likely that the rest of them will realize their true identities in person. Lex will eventually betray the Super Villain Girls.
This will result in the Super Villain Girls getting even with Lex for backstabbing them, possibly even teaming up with the Super Hero Girls to defeat Lex. There will be a Sequel Hook on DCSHG and their 2015 Counterparts
Since the direct-to-video film is going to be having various DC Variants of themselves, with the direct-to-video having mid-credits-scene, with the DCSHG’s 2015 counterparts appearing as a cameo. There will be a Sequel Hook for future DCSHG episodes in The Stinger.
If there are more DCSHG episodes after the movie, maybe they could tease some villains who could serve as an Arc Villain for the second half of Season 2 or Season 3, like Brainiac or Circe. Post-Release
The DC Multiverse will happen at some point in the future
Since it was only focused on the DC Super Hero Girls, it is likely we will have a real focus on it in the future. Sequel Speculations
