- Pretty much anything involving the Bunny Suit is good for a laugh. Your family orders you to take it off and the crusaders are uncomfortable by it (except Polnareff, who likes it a bit too much).
- Joseph's reaction after the Fortune Teller in the Fishing Village predicts that he has an illegitimate son.Fortune Teller: Hmm...Yes...I see it now...I can see your future...I see...A son!Protagonist: A son? Does Miss Holly have a brother...?Joseph: Huh?! Uh, no, er...W-why you! You fraud! Watch what you say!Fortune Teller: Huh? But my predictions are always...Joseph: Shut up! Just take the money and get outta here! La-la-la...
- Even more hilarious if Josuke is in the party. He just glares at Joseph, who's oblivious about what's going on.
- Iggy's Ending, where he basically goes to live with the Protagonist, whether he/she wants it or not. It's even more hilarious in The Stinger, where Iggy gets the poor Protagonist in trouble with his hijinks.
- In one of them, the Protagonist tries to take Iggy back to Joseph and the Speedwagon Foundation. Joseph pretends to lose his phone connection just to get rid of the dog.Protagonist: Well, to make a long story short, Iggy snuck into my bag and came home with me...Joseph: Wow, you don't say! I couldn't find him anywhere, so I assumed he'd run away! Well, maybe I shouldn't say this...But the Foundation had such a hard time with him that I didn't really mind! Haha!Protagonist: Well, about that...Do you think you could get them to take him back?Joseph: Huh? ........Yes. Mm-hmm...Well...*ahem* Take him back, you say...Hello? Can you still hear me? Darn these international calls...Hello? I think you're breaking up! *hangs phone*Protagonist: ....

- In the Chaos Mode, there's a chance to persuade Midler into freeing the Crusaders. If they fail to persuade her and the protagonist is a female Jotaro fangirl, the Protagonist falls into a jealous tirade and starts ranting about how cool Jotaro is. All while Midler is dumbfounded at her screwed priorities and Jotaro is visibly cringing.
- Protagonist: Saying such a thing after Jojo seduced you...Who the hell do you think you are!? I'm not going out on a dud! I've risked my life to travel with him on this journey, and he never said anything like that to me!Midler: That's your problem? I'm gonna eat you and your friends you know...
- What's more, there's a chance to convince Midler with that tirade!
- And you lose friendship points with Jotaro, who's understandably repulsed.
- The Sanctuary Bonus Room has many gems.
- Most of the villains you meet again in Sanctuary are more annoyed that you beat them than remorseful of the rotten things they did that got them killed.
- Amber and Kate (and Midler and Mariah too, if you killed them in the Chaos Mode) crying over being separated from Dio and fighting over him like a bunch of groupies.
- Forever ogling at Dio's female minions.
- Alessi lamenting that he died a virgin.
- Kenny G complains that he doesn't have any custom graphics, instead having the default "defeated sprite".
- The fact that Pearla and Weather Report don't get along in the timeline where the latter wasn't switched at birth, while in the canon universe they were a couple and unaware of their blood ties.Domenico: Man, Pearla...Just get a boyfriend already! Then maybe you'll grow up a little...
- In the Dev Room, Berlin gets accused by the Protagonist of being a pedophile due to his girlfriend's soul residing in the body of a little girl. He's not amused.
