  • Grunkle Stan dispensing some wisdom about aging:
    Stan: Did you find that flying log goblin?
    Dipper: Not exactly. Have you ever encountered black, ominous mystery fog?
    Stan: Sure, every time I take off my glasses! Never get old kids.
  • This exchange, taking the piss out of the What Kind of Lame Power Is Heart, Anyway? trope:
    Jeff: There are four Gemulets, and they are the source of the Gnome Kingdom's magic. The Gemulets harness the magical essences of EARTH, FIRE, WATER and WIND.
    Mabel: Oooh, what about heart? Is there a heart stone?
    Jeff: Please don't belittle our culture. This is serious.
  • The Running Gag about Jeff trying to write off his fellow gnomes' obvious contempt for him as just good old fashioned ribbing.
  • Dipper and Mabel using Schmebulock as a punching bag to test their weapons. Not just the act of beating on the little guy, but the fact that he doesn't seem to mind and that Jeff actively encourages it. There's also him vomiting up rainbows afterward again.
  • This line, delivered by Jeff after the twins return with the Wind stone:
    Oh thank Heavens! Without the Gemulet's power, the forest dies a little each day! The lakes have been drying up, and the birds have been tweeting really hurtful things about my mom.
  • While most of the funniest jokes go to Jeff and Grunkle Stan, the Fire Beast certainly leaves an impression. Though he only appears twice (and only in his second appearance does he actually say anything), and yet he manages to get some of the funniest moments in the game.
    The Fire Beast: HALT! I'm not here for vengeance, Dipper.
    Dipper: You're not? Even after we beat you up and ripped a magic stone from your chest cavity?
    The Fire Beast: Gonna be honest. Didn't LOVE that.
    The Fire Beast: How DARE you! It is NOTHING like a 90's animated movie! Just because you said that, you don't get to hear my song.
    • The Fire Beast denying Dipper's request for help in fighting Dark Jeff on the grounds that he's completely weak without the Fire Gemulet's power. Never mind that even in his depowered form, he's still a massive, hulking minotaur. There's also something inherently funny about the mental image of him playing board games while he waits for this whole thing to blow over.
