Diana’s uncle (or aunt or grandparents, but let’s go with uncle, and let’s call him Ian) was the Chairman of the ICA, inheriting the position from his father. Ian and his brother Peter had some conflict; perhaps Peter had moral reservations, perhaps Ian wanted Diana to succeed him at the ICA, perhaps Peter was planning to usurp him. Either way, Ian ordered 47 to assassinate Peter; Nancy may or may not have been collateral damage.
Ian planned subtly recruit Diana into the Agency, but she fucked up and had to go on the run, forcing Soders to save her ass. That said, she was a stellar handler, and Ian used his position as Chairman to protect her (as said in the books), planning for her to one day succeed him and become Chairwoman.
Unfortunately, that plan went to shit when she betrayed the ICA and leaked his identity out.
