Perhaps, but if that's all it is I doubt Misuzu would be losing sleep over it while staring at Tomo's sleeping face. It is hard to say, though - if she was, steering Tomo toward breaking through Jun's intentional denial would be foolish at worst and some kind of gambit at best, but if she isn't, then she's more or less already achieved her goal, since she's already spending lots of time with Tomo and the others.
What we know:
- Misuzu is intentionally pushing Tomo toward Jun, helping her break through his denial, and pushing for them to have time alone.
- Something about the current arrangement of their group dissatisfies her.
- What Carol says to Misuzu on the road (in #491) is 'The way Jun and Tomo are right now, they may be able to start going out like a normal girl and boy [emphasis present in the original], but I wonder if that's really okay', which clearly bothers Misuzu. Carol has pretty much always been the truth-teller in the group and in the series, and whatever she's observed clearly gets right to the heart of what's driving Misuzu's sudden strange push for Tomo to get together with Jun.
- Strip #496 is Misuzu's inner monologue, while she's staring at a sleeping Tomo. The strip is titled 'I'll Let You Date [Him], So...' with the sense that Misuzu is doing Tomo a favor by letting or allowing her to date Jun (by giving her advice). Misuzu told her to 'be more girly', which she calls 'casual, everyday advice'. Tomo got 'surprisingly' into it, and started spending more time with Misuzu doing girly pastimes that she had never paid attention to before. Misuzu didn't expect that outcome from her statement.
- Misuzu links this to what Carol said to her, and admits that she may indeed be wrong/in the wrong, before thinking that there was something she should have told Tomo instead at that point, as her best friend. She doesn't finish the statement.
- She then says 'but I have no intention of turning back now' - not that it's too late to, just that she doesn't intend to. Here's where the vagueness of Japanese kicks in, though, because she says that by using 'that man', she can be 'more... with Tomo' with the important word left off.
The important part here is that Misuzu's worry, or at least the focus of her monologue - the thing she links Carol's musing-out-loud, as well as the thing she 'should have said as Tomo's best friend', to - is her relationship with Tomo, probably their degree of closeness or intimacy. Something about what she has now is not enough for Misuzu, whether it's more of Tomo's time or Tomo's love, and yet she's actively working against both.
Edit 2: I think Jun's recent flagging on the idea is that he doesn't want to lose Tomo as a best friend by gaining her as a girlfriend, which is consistent with how he's been the rest of the series. My current prediction is that Misuzu is hoping that Tomo will come on too girly, and Jun will 'realize' that he doesn't want her as a girlfriend at all. That's playing a dangerous assumption, Misuzu.

I feel like the first impulse for most people in that situation would have been to check on the girls themselves, and see if they're dented up, or need medical attention? It's awesome seeing the sweet guy go rage, but I'mma just sayin.

I honestly doubt that Misuzu's romantically attracted to Tomo. She's likely being clingy because Tomo's like her oldest friend, and possibly only friend (she clearly doesn't consider Carol a friend), and she doesn't want to spend less time with Tomo because the latter got a boyfriend.
