Starting a Pathfinders game. I've never DM'd before. Any advice?

My friends, some of them experienced players, want me to DM a Pathfinders game. I've played Rifts and done some LARP, so I'm not completely without RPG experience, but I've never led a game before. They're convinced I'll be great at it because I'm a good storyteller, but I'm not as confident as they are. What advice would y'all have for a new DM? As in, how much of the system do I need to know beforehand? How much of the story should I have figured out when we start, and how far ahead of the players do I need to be? How about contingencies? That sort of thing.
