When he fires a Brahmastra at the Pandavas and Arjun fires one back in response, the entire universe will be destroyed unless both of them retract their Brahmastras. Arjun knows how to retract his Brahmastra and does so. Ashwathama cannot retract his - so he targets it at the fetus of Arjun’s grandson. He is cursed with AFateWorseThanDeath as a result.
