Not sure how to categorize this as it might be Real Life, live action TV, or TV Drama.
In the Eighth season of the Amazing Race, on the 12th Leg at the CN Tower in Toranto, Canada; One of the teams chats with the elevator operator, asking if he likes his job. His reply? "It has it's ups and downs."
Might also be I Always Wanted to Say That and/or Incredibly Lame Pun

Seems like there are really two different tropes here: a joke that the characters put a lot of set-up into, versus a joke that the writers put a lot of set-up into. I'm not suggesting creating a new trope or anything but it might be a good idea to split off the in-universe examples into a separate section.
