Negi would be a great tactical warrior king, could lead the people and set up sound policies, but he'd also be too lenient and he'd put himself in harms way too easily. Bad policy for a leader. Asuna... would make a good figurehead, I suppose. She might have some use leading people on the battlefield but then again she'd probably insist on brawling herself.
Asuna is an idiot the same way most idiot heroes are.
She's nowhere near close to ruler material. And, as a figurehead, she is not a particularly charismatic person, but her attitude should earn her quite a few followers.
Both would need advisors, either way, but the difference would be in that Negi can actually act the part of a royal, being a natural born English Gentleman, while Asuna is not going to be a 'proper lady' any time soon.
The thing is, it's not the Megalomesembrian Trusteeship of Ostia. We're talking a kingdom here.

She's older, a hell of a lot wiser, and now she has 400+ years of memories of life in the Royal Family to guide her. Of course she wouldn't be perfect on day 1, but I bet you anything she'd be a competent and well-liked ruled by the time she died.
It's been fun.To be fair, I am judging it basing on Asuna pre-gravekeeper's palace, as I have not seen enough of her to judge.
However, if the scene when she was awaken was anything to go by, she and the Twilight Imperial Princess are separate entities. Which makes sense. Suddenly getting four hundred years of memories, most of which must be of the torture she endured while she was a weapon, all at once should have driven her insane, not calmed her down.
However, I don't think she is unteachable. She is impulsive and temperamental. In a culture that gives a crap about what the president ate for breakfast, any mistake will be pounced upon. And she will have her detractors.
For the record, if I wanted to, I could argue her claim to the throne. It is possible she is not actually related to the royal family at all, since I doubt that she simply appeared one day. Chances are, the Lifemaker created her itself in the case it wanted to blow the crap out of the magic world a boring tuesday. Even if she was, she's part of a branch of the family that has nothing to do with the current royal family, which is composed of only Negi, who if Arika and Nagi never married, as a bastard still gets priority over Asuna.

Simply knowing more about a country's history than anyone else means jack shit when talking politics.
If we really want to talk about a realistic scenario, YES, Asuna would probably be made queen instead of Negi, but she'd probably end up a puppet ruler.

Assuming that Negi actually revealed that he's Arika's son — after all, he could've just limited himself to revealing that he's Nagi's son, since officially nobody that he married Arika on Earth. Alternatively, maybe Negi had abdicated his right to the throne, which makes Asuna the next candidate by simple process of elimination.
