The only game I can think of that I'd like but have absolutely no way to play is Little Big Planet, because it's not available on Steam and there are no PS 3 emulators available yet.
I wouldn't mind trying my hand at Syndicate again, but last time I tried running it in DOS Box it didn't work. Mind you, that was a year or so ago, so maybe the DOS Box devs have fixed enough bugs to make it playable by now?
I'd like to play Homeworld, but I can't even install it. The installation program just crashes on me.
I would like to play some more of Quake 2, but it doesn't want to work under Windows 7. Oh well, at least Quake 3 works.
Pre-XP Windows games seem to be pretty unreliable under Windows 7. Maybe we need something to emulate Windows 98 under current versions of Windows.
Accidental mistakes are forgivable, intentional ones are not.ARMA 2, Stalker: Call of Pripyat, and Fallout: 3 and Fallout: New Vegas on the PC. Big difference from the console versions. >.>
I don't have 2000 dollars to spend on a decent gaming PC.

Half-Life: Dual Nature, a crossover story of reasonably sized proportions.Once upon a time, I got Mass Effect, and it was good. I played it until the part of the game where you first open the starmap on the ship to select your next destination. When I opened it, the game crashed spectacularly, and Google found no solutions.
I really wish I could play Dark Cloud. Disgaea 3, too. I saw it lying in the store way back, but didn't get it when I had the chance. Never saw it again.
Stargate SG-1 Let's Watch. Because my ZHP thing failed.Might and Magic VI-VIII, Heroes of Might and Magic II and III, and Dungeon Keeper 2, due to either having lost the C Ds or having a computer "too powerful" to now play them. :(
And any Zelda game besides Phantom Hourglass and Spirit Tracks, as they're the only games released for DS.
Oh, and Phoenix Wright, as the games aren't available where I live anymore, and the only friend I know who owns them refuses to lend them to me. :/
No More Heroes and its sequel and the Wii version of Sonic Colours, due to my lack of a Wii.
Any particular reason you prefer the DOS Box? Just curious because Syndicate was released for the SNES and there are a few solid emulators right there.
I wouldn't bother with GTA 4. I found it to be a huge disappointment. Oh well, at least I wasn't the one who paid for it.
I have the CD for the DOS version and I like being able to use a mouse. Oh, and the graphics resolution is higher.
Tested it in DOS Box. Still doesn't work. It gets to the point where I can pick either the original or the expansion pack levels, but once I pick one, the game crashes and dumps me back to the prompt.

Live by the sword, live a good long timePortal 2, HL 2 EP 3, F-Zero X (because I don't have an N64), F-Zero (no SNES), and any games newer than 2008, because chances are, my computer won't run it at a playable framerate, playable being above 40.
Also, MAKAI KINGDOMS.
Though with NIS' behaviour these days, I'm just gonna pray for a PSP release.

As for me I'd like to play more Super Robot Wars games. Particularly ones that have non-originals. So far I've played Super Robot Wars 3, Original Generation, Original Generation 2, and Alpha Gaiden.

It's not exactly naive. And it can happen. But it's tough. And definetly worthwhile.
