If it had just been Inside Out 2, I would have thought Universal was trying to blunt Disney's momentum a bit by re-releasing one of last year's biggest hits. But potentially kneecapping their own new release in the process makes no sense to me here.

It’s just a little odd that they’re giving a concrete release date for their Mario Day announcement while being so cagey about everything else.

I'll be curious to see if DK's in the film.
He's not typically part of the Mario cast. He is a vital part of Mario's origin, but he's not a series mainstay like Luigi or Peach or Bowser. He has his own series and he drops in occasioanlly.
However, he was such a big part of the first film. A decent amount of the movie was dedicated to bromancing Mario and DK, which would kind of feel like a waste if DK drops off the face of the franchise going forward. So there's a real possibility he's going to stick around as a part of Mario's supporting cast - At least until a DK-centric spinoff gets fired up, if it ever does.
It's also possible that he'll be given a heavily reduced role just to go "Meanwhile, DK's doing fine," and then quietly swept aside. Just to keep people from asking what happened to DK.

I was fully expecting something like 2028 at the absolute soonest. Though, I guess an animated movie sequel coming out three years after the first movie is natural and makes sense considering Kung Fu Panda 2 came out only three years after the first movie and Despicable Me 2 came out three years after the first movie as well.
Also, can we please stop taking that Miyamoto "Mario characters are a troupe of actors" thing so literally and out of context? He was clearly just comparing them to it or likening them to that kind of thing, he wasn’t straight up confirming that they’re all actors in-universe and that none of the events in any of the games are real.

Maybe they might introduce Bowser Jr. or something.
Like, have the main plot involve Mario and Luigi dealing with Yoshi and trying to raise him, while Bowser breaks free only for the stork to drop Junior off at his castle and Bowser teaches him to be a villain like himself.
You know, parallel character arcs and that.
My guess is a story that incorporates elements of Super Mario World 1 and 2, and a bit of Yoshi's Story.
Mario and Luigi find the Yoshi egg and adventure to find out where it came from and so on. But the more they learn about the Yoshis, the more they discover things they never knew about their own pasts: about how the Yoshis rescued them as babies and brought them back home.
When they reach Yoshi's Island, they then find that the Yoshis of the present are under threat from Kamek / Bowser / etc, so the brothers and Peach have to work together to return the favor and save them.

I wonder if the movie's even going to incorporate the "Yoshis rescued Mario Bros as babies" thing. There's a possibility that they might shift that to Peach.
The first movie made a thing of Peach arriving in the Mushroom Kingdom as a baby, from origins unknown and for reasons unknown. They established that and then left it as a big Question Mark.
If the Yoshi prequel games feature in the Mario 2 film, they may use Yoshi/Stork/Baby Bowser shenanigans to explain how Baby Peach ended up in the Mushroom Kingdom.

Peach, for instance, doesn't boast about never being kidnapped by Bowser, but rather makes it clear that "Fear Becomes Policy" is more her motivation for keeping the hell away from him... and he's in FULL TILT "Great Mighty King Of GUYS WHO TALK TO POSTERS" Mode in this film too, so fair enough.

Don't know why they chose that date as it's 2 weeks after Inside Out 2 and 1 week before Despicable Me 4.
