This may just be how I'm using to doing things, but I think it's best to just do it the 'normal way'. You make a post to start out the RP, but then the players can just post with no restrictions, except when they're doing anything important, or something that has rules, or are talking to an NPC, etc., then the GM comes in to post again. That sort of thing.

On second thought, it does seem a little restrictive. I would like to preserve the turn-based feel somehow, so...maybe an upper limit of three posts between my posts? Or maybe have it only apply during the main story arcs, and have the guide rails off in between for more character interaction?

The system in my 0E RP was basically just rolling initiative per side, having the side that lost declare their actions, having the winning team declare their actions after that, then resolving that round and starting a new one, but... then like you said, there's that problem where someone doesn't post.
As for non-combat.... Well, I suppose that since you're going to have all the NPCs and we'll have our guys, there's always going to be natural points where we can't post anymore (like KK said, talking to an NPC) and will have to wait, so the only problem I can forsee is if everyone runs in on some poor shopkeep with a purchase order, asking where the restroom is, trying to cast a charm spell on him, and the last guy trying to punch him.... then yeah, that'll be a problem

Before we get this show on the road, I'm going to pitch my idea of handling posting in the main thread. Basically, what I may do is have people post in "rounds": that is, I make one post, everyone else makes one post in response to what I post, then I post again, and so it goes. I figure it's the best way to preserve the turn-based aspect of the original game for a forum RP.
Of course, I'm open to criticism, suggestions, and what-not. I know full well how much of a problem it will be if people don't post (as people who have worked with me in the past know), so if you can think of a better idea, I'm up for it.
