I found Toriyama's comment vague. Like, he never makes a connection between Super Saiyan and inheriting S-Cells, and Goku/Vegeta could have passed down their S-Cells before becoming Super Saiyans (if Gohan is being included in this).
It certainly seems to be based on the SS inheritence theory but it's really vague about it.

I really like the idea of the original SSG and SS being enemies. Even assuming the king was waaaay stronger than the rebel Saiyans, wouldn't SSG flatten his SS form?

I was thinking the difference between the two in base would be something truly monumental. Like the low-class Saiyans have Power Levels from 300-500 while the king has a Power Level of like 15,000. The God also would have a solid advantage over the Super Saiyan king, but their massive power level difference in base, as well as the king maybe being a better fighter, lets the king last long enough for the God to run out of time.
On the Super Saiyan God thing, I thought it was saying that the spirit of the first Super Saiyan was looking for someone to become the first Super Saiyan God (for some reason) and not that they were one in the same. Is that not right?
Also, going by the comments in that twitter thread, the name is pun on the word for bean sprout.
That was something I had considered. But if Yamoshi wasn't the Original Super Saiyan God, who was? Or was there even one? Shenron apparently knew about it from the Namekian Book of Legends, so maybe there was no Original Super Saiyan God and the legend is just an exaggeration of Yamoshi going Super Saiyan.

Anyways, I'm a bit disappointed that the original Super Saiyan wasn't called Sadala, I just built that head cannon a month ago. I think Toriyama was stumbling over his words, from the sounds of things, Yamoshi either found super Saiyan on his own, but it wasn't enough to beat the evil Saiyan so he called on the strength of his pure-hearted friends to achieve SSG. Or he became Super Saiyan after his friends died, and after dying himself he sought out his friends to achieve SS God in the afterlife.
And the "S Gene" bit sounds more like a hand wave than an actual lore explanation. I can imagine him shrugging before saying it.
Been playing the Hero coliseum whenever I get enough TP medals for it, the story gives me higher hopes for Xenoverse 3, there are actual dialogue options, Or, atleast, pantomime options. Plus I think the bit with Buu is the first intentional bit of comedy to make me laugh.
Also, Just had this funny thought. Considering Vegeta seems to have a bad track record with gag Manga characters (Neko Majin, Arale, Saitama if you count that.) perhaps the real reason Vegeta completely lost interest in Gohan as a potential replacement rival for Goku was less that he wasn't training and more that he wasn't going to risk losing to the Great Saiyaman because Rule of Funny got in the way of a fair fight.

I'd love a Saiyan arc that reveals that every Universe 6 Saiyan except Cabba is a prodigy in comparison to Universe 7 capable of obtaining bigger and bigger transformations extremely quickly. Resulting in an entire planet of Super Saiyan Gods, while Cabba is still trying to master Super Saiyan 2.

Maybe the Tuffles bust out some sick tech and we get some different kinds of fighters for a change.
It'd be useless against Goku and Vegeta (unless they can create technology equal to God Ki), but with vanilla Super Saiyans, we might get something interesting.

Didn't DB Multiverse have something like that with one of its universes? Can't remember if they were ever stated to be Humans or Tuffles (Cause really the only difference seems to be height and aging, and even then Roshi And Krillin could probably pass for tuffles.)

Man, I even had names for the characters in my version and everything! I was going to name all the low-class Saiyans from 1,000 years ago after extinct species of plants, and the Original Super Saiyan God was going to be named Sagaria in my version. The king was going to be King Saiya the III (his grandfather pulled a Freeza and renamed their species after himself).

I think this confirms that there actually was no Original Super Saiyan God. It also indirectly confirms that living in a gentler environment is why the U6 Saiyans can transform so easily.

It did, but the author made them into a gimmicky OC race called "Heliolites" instead of just making them Tuffles. Not-Tuffles managed to defeat Freeza with their not-Tuffle technology (which admittedly bears no resemblance to anything we saw Raichi or Baby use), and basically became the top of the food chain in their universe.
It was a neat idea - tech-based characters with weapons that could hurt the Z-Fighters, battle suits that were actually war armors. But they mostly get steamrolled throughout the comic and have minimal relevance to the plot. They were squandered. I'd have just made a "Tuffles win and fare better than the Saiyans did," universe.
