This is a big tournament with a lot of fights, which means it's going to be an arc of decent length that - by definition - will have a lot of repetition. Most long tournament arcs focus on an ensemble for that very reason, because in that kind of plot there's only so much a single or even two characters can carry before it just becomes the same thing over and over with slightly different opponents.
For that reason I expect a wide use of the characters and a few major subplots, including some focus mini-arcs for the other members of the team.

I don't think there is. Buu doesn't need to train for it; just see it (he copied the Kamehameha by merely seeing it).
But I'm sure the writers or Toriyama himself will either come up with a reason why that won't happen, or just never bother, so it's a moot point.

As a story, it's actually very short, and the small number of opponents and relatively lower stakes made is easy for Goku and Vegeta to run the show with only minimal input from the other characters.
This story can't really be the same thing because, for one, it would need to be three times as long at least - and that's only if the tournament actually turns out to be single elimination. There are a lot of characters colliding here.
And the more fights and matches there are, there more there needs to be in between those matches to keep the action from oversaturating the story and getting boring. Goku and Vegeta are going to be the protagonists, for sure, but they can't carry this entire thing on their backs like the could the earlier storylines.
Boo still has stamina, can sustain damage, or be instantly obliterated. The Kaio-ken isn't necessarily safe for him even with his advantages.
Furthermore, there's no implication it can be taken up to an infinitely high degree as long as you survive it. We've seen it go up to x20 at most, with no idea how high the ceiling might be.
At any rate, there's no real reason it would be impossible for him to learn it, but Goku's a combat genius and Kaio was still amazed that he could learn it. They don't really address why other characters (like Piccolo) don't have it, but it's probably meant to be because of how difficult it is to learn.
The fact that anyone can potentially learn it is probably why Toriyama retired it, because it'd create problems if every threat and ally needed to have their own Kaio-ken to keep up. At least with Super Saiyan he can choose who has it, and created villains/allies who can keep up with that power in other ways. If Kaio-ken was still around and anyone could learn it, then every villain would need to have it, otherwise the audience would know the villain isn't as strong as they could potentially be. It'd be very monotonous.
