Jenova is a big spooky space virus (technically a prion? whatever, it behaves like a virus so we'll call it a virus) that has been feeding on Planets and whatever unforunate species that happen to be on them. Jenova sends down an "emmissary" of sorts, which takes the shape of whatever native life is present. From there it infects and assimilates both the living and the dead, and manipulates their behaviour via subtle psychological conditioning/viral-parasitic mind control (which is actually a real thing and is really cool to read about). It hides in the bloodstream of its infected thralls, and among the infected populace as perfect replicas, some knowing, some not. It infects both body and soul, so when the infected pass on it infects the Lifestream as well.

Upon reaching the peak of its feeding/reproductive cycle, it triggers the Reunion instinct; since Jenova is technically still one hivemind organism, it will call each and every last bit of itself back to the source, compelling them to gather at one spot, and absorbs the tainted biomass, as well as the remaining unclaimed Lifestream which it feeds off of, into itself, and calls down its true body into the planet's atmosphere, where it rips apart the now lifeless husk of the planet and departs to find something else to eat/reproduce with.

Then some dipshits found it buried in the ice and injected a foetus with it, which brings us to

Mako is the product of processed Lifestream. Said processing removes its connection to the Lifestream and its potential dangers, but simultaneously renders it highly corrosive and incredibly toxic. It is also a known performance enhancer, since it's a processed, chemical form of the source of magic and life itself.

So, why use it in tandem with Jenova? Apart from the fact that both mako and Jenova infusions yield enhanced physical and magical ability by themselves, mako acts as a sort of psychic insulator: the body's cells, now inclined to feed off of the Lifestream, including the body's own soul — bond with the mako instead, which it cannot properly "digest". This prevents Jenova from corrupting the host's DNA any further, as well as offering a performance boost, and Jenova in turn prevents unbonded mako from ravaging the host's cell walls.

Jenova without mako injections present result in bizarre mutations, rare forms of cancer, what can only be described as "wanderlust", memory loss, brain damage, a host of other unpleasant symptoms associated with widespread organ failure. and paranoid delusions of hearing voices that aren't in any language or being watched. Death usually occurs within two months.

Mako without Jenova, on the other hand, is not without its drawbacks — mako is still highly corrosive and equally carcinogenic, if not moreso, due to its potential to become airborne in the form of fumes. As mako still acts as a psychic insulator, this in turn can damage one's psyche considerably, leading to brain damage, violent mood swings, more common forms of cancer, less bizarre but still unpleasant mutations, memory loss, depersonalisation, and other fun side effects that result from having your soul slowly eroded.

As evidenced, it's difficult to tell which symptoms are wholly Jenova's and which are the result of mako poisoning, and using them together does nothing to decrease the risk of any of them — it all kind of balances out, honestly. It's just a matter of what kind of brain damage you want.

The Lifestream is no less potent, and like Jenova possesses a will of its own. It rests at the core of every Planet, and each living thing contains a small part of it, to be returned to it and scrubbed clean upon death. While mako is derived from the Lifestream, it's completely inert to it — the Lifestream is a metaphysical place as much as it is a substance and a creature with a will, and mako is that substance rendered completely inert down to its base chemical properties. While the Lifestream can create more of itself, this requires many, many years, the same way materia is slowly replicated and mastered through one pouring its own knowledge back into the materia.

The Lifestream is capable of weaponising itself and destroying small quantities of Jenova via choking it out/burning it away, but to burn away the infection it requires a significant majority presence — one part Jenova per million parts Lifestream, thereabouts, while Holy can eradicate it outright. These two presences can also operate jointly, though Holy, like Weapons, are merely forces directed by the Lifestream.

The Lifestream is yet another thing that will give you brain damage and erode your soul with the sheer magnitude of it being washed over by the will of the Planet itself and everything that's ever died ever until you lose yourself in the deluge if it doesn't drive you mad first, but at least it won't give you cancer.
