I imagine Vulture will be showing up as part of the Sinister Six, but eventually his sympathetic side will win out and he will undertake a Heel–Face Turn, at which point yadda yadda he dies while saving Spidey and/or ensuring the defeat of the rest of the Six.
The next Big Bad of the MCU will hopefully be Doom or Galactus. Both are long overdue to have some justice done to them.
Edit: The Beyonder might be a dark horse that might actually make sense come to think about it.

I don't think I watched the Morbius trailer. If I did, I totally forgot about it.
I think that exact thing will happen, but with Prowler, not Vulture.
I mean, they could bring back the younger actors from Apocalypse onwards. Give them some good stuff to work with and all.

One Strip!I think MCU Prowler is going to be closer to the original 616 comics Prowler, who tried out two nights of supervillaining, got beat by Spider-man, and then Prowler decided to be a good guy and Spidey's ally from that point onward, even going so far as to dress up as him to cover for Peter Parker.

In terms of grand villains for the MCU, I have some thoughts:
- Dormammu: Strange technically beat him once with the Time Stone, but the Time Stone is no more. As a magical foe, he also provides a different take compared to Thanos.
- Shuma-Gorath: Same as above but with a even more eldritch angle to it.
- Galactus: The cosmic side of the MCU has a lot of room to grow and a entire movie of the Avengers holding off his herald(s) before Galactus shows up in Part 2 could be sick.
- Beyonder: They have adapted the Infinity Gauntlet saga, might as well adapt Secret Wars.
- Masters of Evil/The Thunderbolts: Related to the above point, I think we're at the stage where "evil Avengers" is a pretty easy and fruitful idea that would also give a very interesting upside down spin on the MCU crossover method (i.e we slowly see the villains assembling over the movies instead of the heroes) and an apple to bite for the future director (with a Ensemble Cast of many heroes and villains).
The possibility of Shuma makes me wish Tony was still alive. He would've been perfect as the cocksure man of science who thinks all that elder god stuff is no match for his intellect, delves into terrible eldritch secrets and takes a (temporary) ride on the insanity train.
I guess they could do it with Shuri or Banner, but it just wouldn't feel the same.
The most likely candidates for a new Thanos-level big bad would be Kang, Galactus, the Beyonder (if they're doing Secret Wars (1984)), Doctor Doom, Annihilus (if they're doing Annihilation) or some Thunderbolts/Masters of Evil supervillain group.
People have been saying that it looks like Young Avengers is being built up.
A Thunderbolts/Masters could be a good thing for them to face.
Who claims the role of successor to the Avengers? Sort of deal.
Forever liveblogging the AvengersMostly the comics lineup: Luke Cage, Iron Fist, Spider-Man, Dr. Strange. Jessica Jones was a reserve member in the comic, but they could add her in too. Then add an addition or two from the current MCU, like Shuri, Jen or Kamala. This might be overstuffing it, but they could even introduce Wolverine in one of their movies.
It's interesting, because most of that team were (except Dr. Strange lol) street level characters - but with very hard hitting powers. And all of them are characters who are best described as people with down to earth lives who live within the crazy world that the superheroes created with all its faults, and want to fight to make it a better place - whereas the classic Avengers were a bit more larger-than-life and didn't have that "everyday life" feel to them.
In this "the Avengers saved the universe, and suddenly vanished" world, it's a cool sense of regular people (and Dr. Strange lol) stepping up to fill the void that I think is perfect for the MCU right now. Especially if they do the Thunderbolts, a team that intentionally presents itself as a flashy, patriotic and larger-than-life to fool the people into thinking they're just like the Avengers.

We'll definitely be getting that on a very small scale with Falcon & Winter Soldier, as they'll have John Walker put forth by the US government.
It'll be interesting to see what they do with him-he can easily be played as straight up antagonist, but I imagine he'll most likely end up working with Sam and Bucky in some capacity against Zemo.
I also really want Shuma-Gorath to be the next big MCU Big Bad.
It'll probably be Kang though (friggin' Time Travel bullshit).

I doubt Vulture is coming back as Vulture, but Toomes may show up in some capacity in a later Spidey flick (or big crossover event). Was Shocker ever captured? If he's still on the loose, he totally could pop up later.
I think Gyllenhaal's Mysterio is dead for sure, but a successor or LMD or whatever could be brought in as the plot demands.
