The RX-78-2 is generally larger, at 60 feet vs Optimus usually being portrayed at 40 in his larger incarnation. G1 Optimus Prime is 22 feet according to the original show's scale shart. He's less than half the size.
Honestly, If they stick to G1 Optimus Prime, he's got no chance. He's got both size disadvantage, and he can't fly. Outside of them drawing on some specific feat from the Marvel or IDW comic I can't really think of on the top of my head, I don't see Optimus having much of a chance. Optimus' best asset is likely his experience, but I doubt it'll do much.

Are they using G1 Optimus?
There are so many different versions of him, including the anime ones with Super forms.
It would honestly be impossible to amalgamate them all.

They do have medals for almost, and they're called silver!Well if you go by Japanese Continuity, all the versions of Optimus Except the Beast Wars version are the same guy.
But yeah, if they are going for the strict G1 look, he's fucked.
One point I’ve seen brought up in Optimus’ favor here is that he’s a bio-mechanical lifeform, compared to the Gundam which is a mech suit piloted by a human: meaning that Optimus could act and react to things immediately while there would supposedly be more of a lag time between Amuro’s actions and that of the Gundam.
No idea if any of that true, just passing it along.

The pilot for grandaddy Gundam is a newtype. Which means he has low-key precognition when in combat. Basically, he can start reacting before his opponent actually acts.

What's the RX-78-2 defense against G1 Optimus using the Matrix of Leadership? Are we just accepting that Optimus can't/won't use the Matrix in this battle?
(Actually, it would be a cool result if Optimus used the Matrix of Leadership, and that revealed to both competitors that they were being manipulated into being in a Death Battle by Wiz and Broomstick.)
The Matrix isn't a weapon. It only kills Unicron because he's explicitely vulnerable to it (In the comic because it's a fragment of his antithesis, Primus. In the cartoon, coz it's the computer that helped build him).
On its own the Matrix harmless to anything that isn't Unicron.
In the G1 cartoon, the Matrix is also used to take out a Hate Plague.
Still not a weapon. It still doesn't kill anyone but Unicron. So not seeing your point.
By countering the hate with the goodness inside the Matrix of Leadership. Don't think it will do much against a gundam.
