Her relation to Mr. Spender has not been stated, but given the shared last name and the blonde hair (I almost typed that as "blonde haire"), it's a safe bet that the two are related.
Mothman is one of the two radio hosts from chapter 6. He first showed up here (in poster/vocal form), and here in person. He's not a real mothman, that's just his DJ name.
