The lead in Tsundere seems to imply Tsunderes, by definition, are also Plucky Girl— at least Type A Tsunderes. Does that mean Tsundere is a subtrope of Plucky Girl—or, at least, I don't need to put Plucky Girl in a Tsundere character's trope list for her tsuntsun side?

There is no requirement for a Tsundere to be brave and optimistic. A type A can be snarky and cynical and still count. She's just not a Plucky Girl. In fact, I'd hazard that most of them don't come anywhere near the idea of the Plucky Girl 90% of the time.

Yeah, I'd actually say that Tsundere and Plucky Girl, while not entirely mutually exclusive, are highly unlikely to be seen in the same character.
