I think Shantae might be the most likely third party character to get in as DLC, with Shovel Knight and Bayonetta just behind her.
Anyone else might be harder to think of. Tails and Knuckles would be more likely than most other third party reps at this point (I'm not holding my breath for Snake, Bomberman, or Simon Belmont because of the recent stuff with Konami. And I don't think characters like Travis Touchdown, Sora, or Scorpion are too likely).

De Romanīs, lingua Latina gloriosa non fuī.Of third parties not in Smash, Square and Level 5 are the most likely to get a rep. Square due to their long history and legendary games. But most of the big Final Fantasy characters are too associated with Playstation, which leaves a Dragon Quest character, and I don't know enough about that series. I don't expect a Square character.
I do expect a Level 5 character either as Smash 4's DLC or in Smash 5. Layton and Jibanyan are both very qualified, but also have various things going against them. Layton is a step even below Mega Man and isn't very active. Jibanyan is huge in the most important region, but as of now is untested in the west, though the Yokai Watch anime seems to be having a decent start.
She also boxes gloves in Sonic Battle which makes her equally liable to become a brawler.

I mean, I've warmed up to multiple third-party owned characters, if only because Ryu kinda forced me into it, but I'm gonna stand firm- Think of another Sega character instead of someone from Sonic.
... Maybe go crazy and have it be an Atlus character!

De Romanīs, lingua Latina gloriosa non fuī.I'd thought about something similar years back as well. Basically, take at least one characdter from every game in Sega Smash Pack 2. But, nowadays, we'd need to reconsider what series would be present. Phantasy Star and Shining Force should have several characters (ZYLO, motherfuckers!), despite not being huge series, just because I want Shining Force in, and PS seems like a classic series. Hell, Panzer Dragoon, as long as I'm thinking of series that start with P.
So, what series could be represented?

Shovel Knight is listed with both Smash games, while Chibi-Robo only has Smash U listed in his section. For extra hilarity, Toad is listed is listed as being part of the Super Smash Bros. series when he should be part of the Super Mario series.
The real conclusion to draw from this is that Nintendo should fire whatever intern manages that part of the site.
