It was vague, but he clearly did mean characters who were from a Manga/Anime, which probably meant originating it one, since that's the only way to really decent define them as one, their origins.
Mario has had Anime appearances, same with Sonic. Sonic got in, after all. I really think he meant origins.
But we'll never know, so eh.

I think this is blatant overthinking. With that he meant video game originated characters with games on Nintendo consoles. I don't see that criteria as something not reasonable or something that requires throrough analisis.

Live action movies/television still fit his criteria as okay due to his wording regardless. Also, toys are fine, since he had no qualms about putting a toy originating character in Smash more than once. R.O.B. and even moreso, Mr. Game & Watch. Especially since some don't counts the Game & Watches as video games, as they are actually called Electronic Toys.
So the only legitimate things I can gather properly is "Anime and Manga" or "Animation and Manga/Comic". Anything else? Nope. Rest fall fine under that criteria. Not very vague at all once it got translated, either.

Yeah, I don't get why Electronic Toys aren't always considered Video Games either. But eh, we've seen it even here, so that's a matter of Toy =/= Video Game, respectively so.
Anyway, I'm looking for the wording, but I'm damn sure it was "Anime and Manga", possibly even saying originating. Either way, it clearly does not apply to every media beyond Video Games so we can obviously ignore things like some movies or live action shows and even toys as "barred" no matter what.
I really wish he did just actually say "originated in a video game", but either he didn't mean that(this could be because he considered R.O.B. and Game & Watch toys first?) or he was annoyingly vague(like usual). I'm going to go with "video game origin was not a legitimate criteria, but they clearly need to be in a Nintendo video game to be playable" though, which was an officially stated criteria.
Recently somebody pointed out to me somewhere that the "originating in a video game" was never legitimately said and was a misinterpreted from his actual words. I really don't know how you can get that far unless you actually ignore any media that isn't a Video Game, Animation, or Book, but... here we are.
