I'd want to see someone from Punch Out. Not Little Mac, but one of his opponents. Possibly Super Macho Man...
I like several that have already been mentioned; GLaDOS, Guybrush Threepwood and Spy being my top three.
My suggestion, though? Manny Calavera and/or Glottis.

More importantly though, I'd want to see some sort of online multiplayer mode where you can wager TF 2 items or even steam games.

But overall, I dunno if that would work out.

Sam and Florence from Freefall have been drawn by Mark Stanley playing poker in a non-canon strip (and Sam has appeared playing poker in a couple of canon strips (cheating, I might add)).
Somehow, I can see Jazz Jackrabbit or Roger Wilco working, too.
Accidental mistakes are forgivable, intentional ones are not.It's hard to think of characters with well developed personalities, that have voices, are funny enough, are on the less villainous side, have some footing in the American gaming industry, and are not famous enough to ignore this game.
I could go for seeing Elaine Marley, Ratchet, The Soldier, and Sly Cooper.
But would they still play poker and randomly switch with the existing characters, or play a whole new game? The only other game I think they could play is Bridge.

Never trust anyone who uses "degenerate" as an insult.You know, a simple way to get more life out of the existing game would be to just patch it to add more characters, much like how Valve keeps patching Team Fortress 2 to add new stuff. Still play against four computer opponents at a time, just randomly pick which four characters the player will be up against each time a new game is started.

I kinda think Dante or Nero would like a bit of card playing (especially considering Nero's moves...). I wonder how Heavy Weapons Guy would get along with them.
