  • Awesome Music: This game possesses some really impressive tunes, but special mention should go to the intro and ending themes, "Red Fire" and "It's a Beautiful Day" respectively.
  • Crowning Moment of Heartwarming: Among the various scenes we see in the ending has Elise cradling Isaac with a clear motherly expression. Considering how he's treated by his own mother, it's rather touching.
    • We also have Ballos, seemingly cleansed of the violent anger that consumed him and created the Red Crystal, running a noodle shop and serving food to Astro, Quote and Dr. Black Jack. After the horrors that he'd been privy to and inspired that anger, it's sweet to see him finally able to move on.
  • Epileptic Trees: When it was announced that Peter Cullen was doing voiceover for the intro, people immediately took it as indication that Optimus Prime would be in the game.
  • That One Attack: Most characters with a single Burst Meter have Bursts as powerful as you'd expect.
    • Akuji's Demon Reflection swaps grids with his opponent. Which means he can turn a doomed situation into an instant win.
    • Kawase's "Go Fish!" steals any special crystal on the opponent's grid, including their next drop. About to get a Poly-Crystal and turn the tide? Nope! It's hers now and your bad situation is now even worse.
    • Zombie's Viral Infection in Survival Mode. In this mode your grid isn't cleared after defeating an opponent which means you'll still be dealing with its effects long after he's been taken out.
  • Unexpected Character:
    • Johnny Turbo has got to be one of the most out there guest characters in recent times. note 
    • Astro and Black Jack have been in an obscure Columns puzzler before, but nobody expected Nicalis to license them of all characters for any project.
    • Ballos is a surprising choice considering his existence was only even revealed in the Golden Ending, especially when the Doctor is a non-spoiler option for a Cave Story villain. That said, the Final Boss is the Undead Core, the Doctor's One-Winged Angel form, with the implication that Ballos was working against him.
