This is why he is an old man as a human.

There will be a reference to “Griffin’s redesign as a hottie” joke.
Would they even have the time to put that in?

There will be No Antagonist.
Maybe the film will keep the "heroes trying to reverse their transformations" as the core of the story.

Dracula will turn out to be dying of old age by turning into a human.
Since he was a vampire, he must have lived for so many years. So, becoming mortal may have consequences that should be stopped. Additionally, Dracula could undergo a Disney Death, where he confesses that he's proud of Johnny being part of his family. Thus, it would be a good way to conclude two important character arcs.

Mavis will end up turned human

If Griffin's love interest Crystal also becomes human, there will be a joke about the Double Standard regarding the issue of depicting female nudity in family-friendly media
She was shown fully clothed during her short appearance in Hotel Transylvania 3: Summer Vacation, and given how male nudity has almost always been given a free pass in family films, it's unlikely she'll also be naked in this film if she makes an appearance.

Additionally, if Crystal turns human, she’ll be extremely attractive, giving her and Griffin an Ugly Guy, Hot Wife dynamic.

For bonus points, maybe Griffin will be insecure about this, and there’ll be An Aesop about how looks don’t matter.

If Ericka is hit with the Monsterfication Ray, she’ll grow four octopus-like limbs

Alternatively, she’ll become a witch.

After reversing the effects of the Monsterfication Ray, Dracula will retire and pass on the running of the hotel to Johnny and Mavis
Confirmed, it's one of the main plot points.

Johnny will be mistaken for a Alebrije at the airport in Latin America.
Perhaps it is the reason why people in the Latin American airport are not scared or surprised by the dragon form of Johnny at the airport.

Griffin actually used to be handsome.
However, of course he aged and stopped looking good after so much time of becoming invisible. Up until becoming visible, he thought he still looked the same as when he was young, as shown in the trailer when he says "I'm balding?" The third film even shows he was alive in 1897, showing he's as old as his friends.

Ericka will turn into a vampire.

Abraham Van Helsing's robotic parts will become human and his remaining human parts will become robotic.
Jossed.

The film will end up releasing in theaters after all.
After seeing the success of both Venom and No Time to Die, Sony will backpedal and decide to release the film in theaters anyway, especially given that the Amazon release is starting to look tenuous with the originally announced October 1 release date come and gone. True, both of the above are adult films, but the huge box office makes it clear that the appetite for theatrical releases is there.

This film will discuss how humans have a shorter lifespan than monsters.
Maybe Dracula is just worried that Johnny and Ericka will eventually die, thus leaving him and Mavis widowed.

    Post-Release 
Dennis is immune to the monsterization ray.
While we'll never find out (or at least not yet), Dennis might have some level of immunity towards the Monsterization Ray because he's half-human, half-vampire.
  • Given that he's half-and-half, it could work while not working (that is, the vampire part is turned human, but the human part is also turned vampire, so functionally everything's the same).

Frank remembers being human (consciously or unconsciously).
If Frank had no idea of being anything but a monster, presumably he would have reacted to his human transformation with the same horror that the others show. Instead, he enjoys it, because he somehow knows this is attractive for a human, even if it's hideous for a monster.

Dennis can be turned into a full human or a full monster.
But, should it happen, he'll no longer become half-human and half-monster again.

If Drac and/or Vlad knew about the monsterization ray before Dennis' fangs came out, they'd try to steal it.

The monsterization ray was Van Helsing's Plan B.
If not for his Heel–Face Turn, he would have tried to turn Drac into a human as an alternative to killing him. Not figuring out how to affect all of Drac's monster friends was his main reason for it not being Plan A.
