• Cross-Dressing Voices: Touta Konoe is voiced by voice actresses Yuka Takakura in the original Japanese version and Brittney Karbowski in the English dub. Rina Sato reprises her role as Negi Springfield.
• The Original Darrin: Takehito Koyasu returns from the first Negima series to voice Nagi Springfield after Miyuki Sawashiro voiced him in Negima!? (second season).
• The Other Darrin: In the English dub, everyone besides Greg Ayres (Negi), Luci Christian (Asuna), and Monica Rial (Konoka and Kazumi, though both of them were only cameos) was recast.
• Studio Hop:
  • J.C. Staff handled the animation this time around.
  • The original series was dubbed into English by Funimation. For UQ Holder, it was instead dubbed at Seraphim Digital Studios and licensed by Sentai Filmworks. In a twist of irony, Sentai would eventually snap up the rights to the original Negima! series from Funimation in 2018.
• Trolling Creator: Ken Akamatsu manages to continue his habit of trolling his own publishers, with UQ Holder being a stealth sequel to Negima for over a hundred chapters, with the original cast being seemingly Killed Off for Real... only for that very first page to be a Fake-Out Opening of epic proportions, as later chapters brought back a sizeable proportion of the fan favourites from the original series, culminating in Chapter 129, where it gains a new title... Mahou Sensei Negima 2: UQ Holder.
• What Could Have Been:
  • There were two prototypes written for the series. They were eventually released in manuscript form in Bonus Material. Both share elements of: A destroyed space elevator, and each Touta prototype being named Kikuchiyo.
  • The first is about the young orphan, Kikuchiyo being under the wing of an eyepatch clad Eva. Kikuchiyo is originally shown to be hostile towards Eva, due to seeing her kill a nun he was apparently close to. At the end, Evangeline dies, complete with a Grave-Marking Scene.
  • The second is '7 Samurais', the titular group led by Eva that seems to named after based on Seven Samurai, whose hequarters is based in the slums area with the label Samurai 7. Jinbei, Gengoro, and an older looking Ikkuu appear as members. Also present is a girl who strongly resembles Mizore.
