• The Original Darrin: While the Italian dub of Lupin III: Part 6 replaced all the main cast except for Jigen, here Stefano Onofri, Rodolfo Bianchi and Alessandra Korompay reprise their roles as Lupin, Zenigata and Fujiko.
• The Other Darrin:
  • Rui Kisugi is voiced by Rika Fukami in this film as her original seiyuu Toshiko Fujita passed away in 2018 (her previous appearance in City Hunter: Shinjuku Private Eyes was voiced Keiko Toda, in addition to the latter reprising Hitomi)
  • Mr. Nagaishi is voiced by Mugihito as his original seiyuu Tamio Ōki passed away in 2017.
  • For the English dub, the Cat's Eye sisters are voiced by different actors than who voiced them previously in City Hunter: Shinjuku Private Eyes.
    • Rui Kisugi: Dawn M. Bennett —> Cristina Vee
    • Hitomi Kisugi: Alexis Tipton —> Alicyn Packard
    • Ai Kisugi: Megan Shipman —> Deneen Melody
  • In the Italian dub:
    • Goemon is voiced by Davide Perino instead of Antonio Palumbo.
    • The Cat's Eye characters are also recast compared to both the original series and City Hunter: Shinjuku Private Eyes: Elena Perino replaces Barbara Castracane and Simona Biasetti as Rui, Domitilla D'Amico replaces Claudia Razzi and Giuliana Atepi as Hitomi, Emanuela Ionica replaces Susanna Fassetta and Daniela Fava as Ai and Emanuele Ruzza replaces Teo Bellia as Toshio.
• Role Reprise:
  • The current Japanese casts of both Lupin III and Cat's Eye (with the exceptions of Rui and Nagaishi) return for this film.
  • The current English dub cast of Lupin III since Part 4 also all reprise their roles here.
