- The Original Darrin: After being recast with imitators in the original release, XL brought back the original actors for Blossom (Cathy Cavadini), Numbuh One (Benjamin Diskin), Billy (Richard Steven Horvitz) and Flapjack (Thurop van Orman).
- The Other Darrin: Several characters, at least in the original 3DS version, were played by different actors from the ones they had in the shows they originated from. The updated XL version recast several characters with their original voice actors.
- Yuri Lowenthal replaces Thurop Van Orman as Flapjack, Benjamin Diskin as Numbuh One, David Coburn as Captain Planet, and Greg Cipes as Kevin Levin.
- Grey DeLisle voices Mac instead of Sean Marquette and Blossom instead of Cathy Cavadini.
- Tara Strong replaces Candi Milo as the voice of Cheese and Dexternote , Nicky Jones as the voice of Chowder, Ashley Johnson as the voice of Gwen Tennyson and Elizabeth Daily as the voice of Buttercup.

- Quinton Flynn replaces Richard Steven Horvitz as Billy and Dee Bradley Baker as the Toiletnator.
- Role Reprise: A select few of the original actors reprised their roles for the original release: Yuri Lowenthal as Ben Tennyson, Tara Strong as Bubbles, Keith Ferguson as Bloo and Grey DeLisle as Mandy. The Updated Re-release retained all of the aforementioned actors and also brought back several more reprises: Cathy Cavadini as Blossom, Benjamin Diskin as Numbuh One and Numbuh Two, Richard Steven Horvitz as Billy, Thurop Van Orman as Flapjack and Tara Strong as Young Ben.
- Troubled Production: During the games production the developers worked through 14 hour crunch days as well as using out dated technology such as a physics engine with collision bugs for it's 10 years of use and trying to fix it caused more bugs. Also, Co-op was added late into production by forcefull demand of the publisher.
- What Could Have Been:
- Unused narrator voice clips suggest that Mandark, Shnitzel and the Remote Control were all intended to be playable characters at some point in development.
- Character icons for Stickybeard, Wilt and Gwen have been found in the game files, though what role they may have played is unknown.
