A story about pinmonkeys and burning your bridges. And one of the most heartwarming scenes in the entire series: "do it for her"

Homer The Great

For just being hilarious.

Lisa's Wedding

For giving us the most rounded and touching view of the family in possibly any episode (and appearing to be a bit of a precursor to Futurama).

Lisa's First Word for being both funny and absolutely adorable.

El Viaje Misterioso de Nuestro Jomer (The Mysterious Voyage of Homer)

Oh, there are so many to choose from. I like Homer's Phobia a lot, though.

Round Springfield

"Like when a sign says do not feed the bears then you better not feed the bears"

Bart vs. Australia

The Way We Was

The Way We Was is a wonderful summation of what makes The Simpsons, at their best, one of the greatest families (animated or otherwise) to grace television. Laying the groundwork for Homer and Marge's relationship; we see Marge at her most understanding, Homer at his most repentant and hey: Jon Lovitz!

Holidays of Future Passed

The "just in case" christmas episode, produced as what would've been the last episode of the simpsons before it got contracts ironed out. Another future episode, seeing the future trajectory of all the major characters of Springfield and how the world changed around them. Also a ball of family and Christmas cheer that would've made for a good sendoff too.

22 Short Flims About Springfield

Steamed hams, free nose jobs, stuporous funkers, big guys in tiny cars, steamed hams, babies trapped inside newspaper boxes, woodpeckeros, McOnal's burgers and of course... steamed hams.

It's difficult to sift through 500+ episodes of animated history and say definitively which episode is the best. But Simpsons Roasting On An Open Fire, the series first episode, is without a doubt an important touchstone for the series. If not the best episode, certainly one of the best holiday themed pieces of animation, Simpsons or not.
