IIRC, Poison's gender was introduced because of sexism. Somebody in the development process didn't like the idea of having the male player characters beating up women and a Designated Girl Fight wasn't on the table, so having the player beat up transsexuals was introduced as a solution.
It's a common, heavily condescending misinterpretation of the "Do not hit girls," social value that devalues female combatants by declaring them off-limits no matter how capable they are.
It has also turned Poison into a social minefield of a character, because if you insist on the trans interpretation, you are contributing to the "Women can never be fighters," mindset, and if you insist on the female interpretation, you are contributing to trans erasure.
All things considered, Manly Comics has handled this remarkably well and managed to avoid both pitfalls by having other badass female characters in the comic and using the trans portrayal to make a point about tolerance.

The sad, REAL American dichotomyI mean, yes. That's where the idea originated from and that sucks, but if Poison is trans she is still a woman. She isn't a man dressed up as a woman. And she is a fighter. So, really, their entire plan there just kinda didn't work at all.

Oh really when?I believe a lot of these were rumours. This video (in the first 3 minutes or so) claims that she was always supposed to be trans, and was indeed replaced by a man at one brief point, but she wasn't changed from female to trans.
